Antimicrobial resistance and genotyping of Mycobacterium leprae in Venezuela

چکیده
Leprosy is a neglected tropical disease that leads to physical disability and social discrimination. The active surveillance of new cases vigilance for drug resistance can decrease the incidence, improve clinical outcomes people affected by it. We analyzed, with molecular biology techniques, set skin biopsy samples from 25 Venezuelan patients. patients had been diagnosed leprosy in 2014 early 2015, 15 were relapse cases. tested dapsone, rifampicin, fluoroquinolones. In addition, we performed epidemiology analysis through multiple-locus variable-number tandem repeat (MLVA) single nucleotide polymorphism (SNP) typing. did not find evidence 76% ( n = 16) isolates belonging SNP type 3. Genotype profiles allowed us rule out possibility re-infection patient persistent symptoms after treatment, as well household transmission two more Although our sampling relatively small, very similar or even identical Mycobacterium leprae genotypes observed Miranda State. presence this cluster highly suggestive high rates local and, turn, need better control disease. Finally, copy number distribution minisatellite 18–8 considerable 3 strains strongly suggests sublineage particular Venezuela.

Dapsone resistance in Mycobacterium leprae.
The folP1 gene of Mycobacterium leprae, which encodes dihydropteroate synthase (DHPS), was studied for the presence of mutations associated with resistance to dapsone (DDS). When the folP1 of several DDS-resistant clinical isolates of M. leprae were sequenced, two missense mutations were identified. One mutation occurred at codon 53, substituting isoleucine for threonine in DHPS-1, and a second...
متن کاملCharacterisation of short tandem repeats for genotyping Mycobacterium leprae.
OBJECTIVE Establish a typing system for Mycobacterium leprae based on polymorphic DNA structures known as short tandem repeats (STR). DESIGN Assess 16 polymorphic STR for sensitivity, specificity and reproducibility in standard assays using reference strains of M. leprae. RESULTS Primers for 16 STR loci were selected based on PCR product size and for their ability to sequence each STR locus...
متن کاملMolecular basis of rifampin resistance in Mycobacterium leprae.
Rifampin is currently the most potent drug used in leprosy control programs. We show that the rifampin resistance which emerged in nine patients with lepromatous leprosy, who had received rifampin monotherapy, stemmed from mutations in the rpoB gene, which encodes the beta subunit of RNA polymerase of Mycobacterium leprae. In eight cases missense mutations were found to affect a serine residue,...
متن کاملDihydropteroate synthase of Mycobacterium leprae and dapsone resistance.
Two Mycobacterium leprae genes, folP1 and folP2, encoding putative dihydropteroate synthases (DHPS), were studied for enzymatic activity and for the presence of mutations associated with dapsone resistance. Each gene was cloned and expressed in a folP knockout mutant of Escherichia coli (C600DeltafolP::Km(r)). Expression of M. leprae folP1 in C600DeltafolP::Km(r) conferred growth on a folate-de...
متن کاملGenotyping of Mycobacterium leprae for better understanding of leprosy transmission in Fortaleza, Northeastern Brazil
Leprosy is endemic in large part of Brazil with 28,761 new patients in 2015, the second largest number worldwide and reaches 9/10.000 in highly endemic regions and 2.7/10.000 in the city of Fortaleza, Ceará, Northeast Brazil. For better understanding of risk factors for leprosy transmission, we conducted an epidemiologic study supplemented by 17 locus VNTR and SNP 1-4 typing of Mycobacterium le...
